This comprehensive review explores the critical role of Transforming Growth Factor-beta (TGF-β) signaling in the pathogenesis and progression of gastrointestinal and hepatic diseases, including cancer. TGF-β is a key regulatory molecule influencing cellular growth, differentiation, and immune responses, often altered in cancer. Genetic mutations affecting the TGF-β signaling pathway are prevalent across various cancers, particularly those of the gastrointestinal tract, liver, and pancreas. The pathway, mediated by SMAD proteins, has dual roles, acting as a tumor suppressor in early stages and potentially facilitating cancer progression in later stages by modifying the tumor microenvironment.
